Retrospective MicroscopyThe creation of histological slices is a common approach
to investigate
tissue and tissue changes on a cellular basis. Sometimes,
however, it
would be beneficial if the underlying three-dimensional
structure might
be viewed as well, for example to be better able to
identify
morphological characteristics. The aim of this project is the 3-D reconstruction of a
set of
histological slices, which includes investigation and
implementation of
the following steps:
Selection of slices suitable for reconstruction
Reordering slices to restore the proper sequence
Artifact reduction
Rigid and non-rigid registration of the slices
Segmentation of Regions of Interest
Saving and displaying a 3-D volume
This project is primarily focused on the reconstruction
of the optical
nerve head. It is of special interest in the case of eye
diseases like
glaucoma. By reconstruction, structural changes in that
region should be
perceived and investigated faster and easier. Furthermore, the possible benefits of a combination of
histological
reconstructions with acquired volumes of other imaging
modalities, e.g.,
