Role of Vinculin in Carcinoma Cells

Among the most common and devastating forms of tumors is breast cancer. Vinculin is known to be a component of the adhesion complex of cytoskeletal proteins in tissue. The disruption and restoration of the interactions of this protein in F9 and MEF carcinoma cells present an ideal opportunity to study the underlying mechanisms of actin-membrane attachments. In the proposed study, we will examine F9 and MEF carcinoma cells deficient of vinculin, produced genetically by homologous recombination, and vinculin-transfected F9 cell lines to clarify both the function of vinculin in cytoskeletal organization and the effect of vinculin on the mechanical and regulatory properties of the cell.
